Differences in the Detoxification Metabolism between Two clonal Lineages of the Aphid Myzus persicae (Sulzer) (Hemiptera: Aphididae) Reared on Tobacco (Nicotiana tabacum L.) Chilean J. Agric. Res.
Cabrera-Brandt,Marco A; Fuentes-Contreras,Eduardo; Figueroa,Christian C.
Myzus persicae (Sulzer) is a highly polyphagous aphid species, with a subspecies (M. persicae nicotianae) well adapted to tobacco (Nicotiana tabacum L.). We evaluated the effect of this host plant on the aphid performance and detoxification enzymes, in order to test the participation of xenobiotic metabolism on the ability of this aphid to overcome the tobacco chemical defences. Two genotypes, one corresponding to the only M. persicae nicotianae genotype reported in Chile on tobacco, and one genotype belonging to M. persicae sensu stricto were reared on tobacco and pepper (Capsicum annuum L.), respectively. M. persicae nicotianae showed a significantly higher intrinsic rate of increase (r m) on pepper than on tobacco, and M. persicae s.s. performed...

Effect of phenobarbital on inducing insecticide tolerance and esterase changes in Aedes aegypti (Diptera: Culicidae) Genet. Mol. Biol.
Sousa-Polezzi,Rita de Cássia; Bicudo,Hermione Elly Melara de Campos.
The effect of phenobarbital (PB) on the induction of tolerance to the organophosphorous insecticide temephos (TE) was investigated in Aedes aegypti L4 larvae submitted to two different PB-treatments:(1) continuous treatment from the egg to the larval L4 stage and (2) discontinuous treatment in which L4 larvae were exposed for 30 h. Mosquitoes from two Brazilian cities were studied: São José do Rio Preto (SJ) in São Paulo State and Goiânia (GO) in Goiás State. According to criterions established by World Health Organization (WHO) mosquitoes from SJ are organophosphate-susceptible while mosquitoes from GO are organophosphate-resistant. For both SJ and GO larvae the two different PB-treatments resulted in significantly increased tolerance (measured by reduced...

Effects of caffeine and used coffee grounds on biological features of Aedes aegypti (Diptera, Culicidae) and their possible use in alternative control Genet. Mol. Biol.
Laranja,Alessandra Theodoro; Manzatto,Antonio José; Campos Bicudo,Hermione Elly Melara de.
Caffeine and used coffee grounds completely blocked the development of Aedes aegypti in the early stages, in treatments with the concentrations 1.0 mg/mL and 50 mg/mL, respectively. More advanced stages and even adults were obtained in lower concentrations of both substances, enabling observations to be made of mortality rate, longevity and esterase patterns. The experiments involved treatments using either eggs or 3rd instar larvae (L3), with or without the addition of fish food. Mortality rates prior to the adult stage and adult longevity were significantly different in the comparisons among treatments, in every kind of experiment, but in those using L3 larvae, their percentages were smaller. Observations of the time of larva and adult onset suggested...

Esterase polymorphism marking cultivars of Manihot esculenta, Crantz BABT
Resende,Adriana Gazoli; Vidigal Filho,Pedro Soares; Machado,Maria de Fátima P. S..
Esterase isozymes were used to detected substrate-preference polymorphism in twenty cultivars of Manihot esculenta, and to show cultivar-specific variation of this species. A relatively complex extraction solution of proteins from leaves was needed to show a larger number of esterase isozymes. Similarity between cultivars from six groups ranged from 51 to 96%. The cultivars identified by the same name seemed to be biochemically different regarding esterase isozymes. Esterase isozyme electrophoretic patterns could, therefore, be used to discriminate the cultivars identified by the same name, and to monitor the vegetative propagation of cultivars maintained in the germplasm collection. In breeding strategies, isoesterase analysis could be used to avoid...

Esterase profile in a pyrethroid-resistant Brazilian strain of the cattle tick Boophilus microplus (Acari, Ixodidae) Genet. Mol. Biol.
Baffi,Milla Alves; Pereira,Cícero Donizete; Souza,Guilherme Rocha Lino de; Bonetti,Ana Maria; Ceron,Carlos Roberto; Gourlart,Luiz Ricardo.
The cattle tick Boophilus microplus causes great damage in livestock and is considered one of the most important tropical ectoparasites. The traditional method of control is based on the intensive use of pesticides, however the indiscriminate use of these compounds over the years has led to the selection of resistant ticks. Hydrolases, particularly esterases (EST), have been reported to be associated with acaricide resistance in B. microplus. We compared the esterase profile of susceptible and cypermethrin-resistant strains of adult B. microplus and a pyrethroid susceptible reference strain (the Mozzo strain) using polyacrylamide gel electrophoresis and specific staining. The electrophoretic profiles of protein extracts revealed the presence of four...

Histology and histochemistry of the venom apparatus of the black scorpion Androctonus crassicauda (Olivier, 1807) (Scorpiones: Buthidae) J. Venom. Anim. Toxins incl. Trop. Dis.
Jarrar,BM; Al-Rowaily,MA.
The venom apparatus of the black scorpion Androctonus crassicauda has been characterized histologically and histochemically in the present study. The results showed that this apparatus consists of paired venom glands, each of which initially presents its own canal and posteriorily both fuse into a single common one. Each gland is covered by a sheath of striated muscle and is lined with extensively folded secretory epithelium (formed of non-secretory and secretory venom-producing cells). The outcomes also revealed that the venom-producing cells of both glands produce neutral mucosubstances, sialomucins, sulfomucins and proteins, but are devoid of glycogen. Cysteine, tyrosine, tryptophan and arginine were also detected along with activities of acid and...

LEAF-CUTTING ANTS Acromyrmex niger SMITH, 1858(HYMENOPTERA; FORMICIDAE) USED AS BIOINDICATORS OF AGROTOXICS RESIDUES Acta biol.Colomb.
BELIZÁRIO CANTAGALLI,LIRIANA; ALVES LOPES,DENISE; PAZ BARATEIRO STUCHI,ANA LÚCIA; COLLA RUVOLO-TAKASUSUKI,MARIA CLAUDIA.
Despite the condition of leaf-cutting ant pests in agroecosystems, it is undeniable the benefits they can bring in certain situations or environments. The leaf-cutting ants of the genus Acromyrmex attack mainly leaves of vegetables and fruit trees exposing not only to the agrochemicals used for their control as well as to those used for the control of other pests. Due to the bioindicator potential of environmental quality of the ants and their frequent exposure to agrochemicals such as organophosphates, neonicotinoids and growth regulators insecticide used for pest control, it is necessary to study the sublethal effects that these pesticides may cause. The electrophoresis technique was used to study the activity of esterase isozymes involved in the...

Low malathion concentrations influence metabolism in Chironomus sancticaroli (Diptera, Chironomidae) in acute and chronic toxicity tests Rev. Bras. entomol.
Rebechi,Débora; Richardi,Vinicius S.; Vicentini,Maiara; Guiloski,Izonete C.; Assis,Helena C. Silva de; Navarro-Silva,Mário A..
Low malathion concentrations influence metabolism in Chironomus sancticaroli (Diptera, Chironomidae) in acute and chronic toxicity tests. Organophosphate compounds are used in agro-systems, and in programs to control pathogen vectors. Because they are continuously applied, organophosphates often reach water sources and may have an impact on aquatic life. The effects of acute and chronic exposure to the organophosphate insecticide malathion on the midge Chironomus sancticaroli are evaluated. To that end, three biochemical biomarkers, acetylcholinesterase (AChE), alpha (EST-α) and beta (EST-β) esterase were used. Acute bioassays with five concentrations of malathion, and chronic bioassays with two concentrations of malathion were carried out. In the acute...

Molecular weight estimation of esterase isoenzymes in closely related Drosophila Species (Diptera: Drosophilidae) in non-denaturing polyacrylamide gel electrophoresis BABT
Mateus,Rogério Pincela; Cabral,Hamilton; Bonilla-Rodriguez,Gustavo Orlando; Ceron,Carlos Roberto.
A method that allows the measure of molecular weight of two well-known and closely related esterases from Drosophila mojavensis and its sibling species, D. arizonae, is here described, using native polyacrylamide gel electrophoresis at several concentrations, applying Fergunson´s principles. These enzymes, namely EST-4 and EST-5, presented molecular weight values between 81 and 91 kDa. In spite of their distinct expression pattern through the insect's life cycle, they showed properties of isoenzymes codified by distinct structural genes, supporting the hypothesis of a rather recent gene duplication event that generated both in D. mojavensis and D. arizonae, as well as in other species of repleta group. The method is simple and adequate to be applied to...

Sex-dependent differences in the activities of acetylsalicylic acid-esterases in mouse kidneys BJMBR
Benedito,M.A.C..
Acetylsalicylic acid (ASA), the most used drug worldwide, is hydrolyzed to salicylic acid and acetate by esterases present in tissues of several species including humans. Sex differences in drug metabolism by rodent liver are documented in the literature. In this paper we report a difference in the activities of the esterases (ASA-esterase I and II) in the kidneys of male and female mice. In this species there is no difference between males and females in liver ASA-esterases (ASA-esterase I: males 38.5 ± 7.9 (N = 5) and females 31.6 ± 7.6 (N = 5) nmol of salicylic acid formed min-1 mg protein-1, P>0.05; ASA-esterase II: males 77.3 ± 17.4 (N = 5) and females 61.4 ± 15.1 (N = 5) nmol of salicylic acid formed min-1 mg protein-1, P>0.05)....
